We're an engineering focused team out of UC Berkeley that has a diverse background from informatics to big data. We've just recently raised a size-able seed round and found ourselves in a beautiful office right in downtown Berkeley. Our stack is JS front to back. We run Node on the backend and React/Flux on the frontend. We build fast, release features every week, and love giving stuff back to the community through open source projects like our player (https://github.com/ToneDen/toneden-sdk).

More specifically, we are able to automate combinatorial gene assembly straight from a web app interfacing with a TECAN microfluidic liquid handling robot.
Here's a proof of concept with a simulator, but I've tested this in a live enviornment.

Functionality for including tracks locally on the server is there also, but we need to modify the API for this a little.
The repeat button is on our dev list. Should be out in a week or so.
The second issue is something we are very familiar with. What we think is happening here is that if the track is buffering too slow and the playing progress catches up with the buffer, our player should just stop playing until the track loads. We are playing with the thresholds to make the player pause before it gets to that case. We are pushing a fix for this soon.
We have the functionality for it there in the source but we just need to implement a UI for it.

The challenge is creating a community for both the musicians and fans to interact meaningfully. Musicians want to be free from being lumped in with the noise and fans want to be explore that content freely. I think for most streaming services out there, this hasn't really been totally figured out, although Spotify and SoundCloud are trying to.
