From the paper's abstract

"We found a very weak or no correlation between AlphaFold output metrics and change of protein stability or fluorescence. Our results imply that AlphaFold may not be immediately applied to other problems or applications in protein folding."

This seems like the paper is drawing the opposite conclusion of the current HN title ("AlphaFold predicts impact of single mutations on protein stability and function") perhaps a title edit is in order?

The fact that AlphaFold is not able to predict effects of single mutations is well known in protein research community. For SOTA in this space, see Deepsequence (https://doi.org/10.1101/235655) and ESM (https://doi.org/10.1101/622803)

This post appears to have a misleading title. AlphaFold didn’t do any prediction, rather, researchers compared its output values with/without single mutations. As far as how to evaluate the success of AlphaFold (from the abstract): “We found a very weak or no correlation between AlphaFold output metrics and change of protein stability or fluorescence. Our results imply that AlphaFold may not be immediately applied to other problems or applications in protein folding.”
