Currently not it seems. It doesn't seem to matter from where I call videoElement.webkitEnterFullscreen(), it always works (in Safari)

I'm not sure of the html5 limitations, but I do know that Flash only allows you to enter fullscreen on a keyboard or mouse click event, so I'd hope the browsers copy this requirement.
